DMS Method
Seawater samples collected by CTD+Rosset system were filtered onto 25mm Whatman GF/F filters using
gentle suction of 150mmHg. After filtration, the filters were dipped in 6ml of N,N-dimethylformamide
(DMF) for extracting chlorophyll a (Chl a). Extractions of chl-a were done in a dark place at -80deg;C.
Chl a concentrations were measured with Turner Designs Model 10-AU Field Fluorometer (Parsons et al.
, 1984).
Acetone Method
Seawater samples for chlorophyll a (Chl a) were filtered through Whatman GF/F filters (Ø47mm, 0.7µm
nominal porosity) at -100 mg Hg vacuum pressure. Filters were folded in aluminum foil and stored at
-20°C in a desiccator until analysis. Volumes of samples varied from 200 to 500 ml depending on ambient
Chl a concentrations. The filters were placed in test tubes containing 5 ml of 90% aqueous acetone.
Extraction took place in the dark at -20°C for 24 h. Chl a concentrations were measured ashore by a
Turner Designs Model 10-AU Field Fluorometer following the methods of Parsons et al. (1984).
