In 1996, the list of states that have experiences natural resource, public health and economic impacts related to this organism expanded, with the addition of Alabama, Mississippi, and Louisiana, to include all the Gulf-coast states and North Carolina. Estimates of economic impacts to Florida and North Carolina from two moderate intensity blooms ranged from 15 to 25 million dollars respectively. The harmful impacts caused by K. brevis occur only when cell concentrations increase significantly above low background concentrations that are present year-round in the Eastern Gulf of Mexico. Once a bloom has developed offshore in typically oligotrophic waters, cell concentrations at the 105 level can be maintained for months. During 21 of the past 22 years, red tide blooms have been observed within the region between Tampa Bay and Charlotte Harbor. The key to understanding any HAB lies in knowing how one algal species has adapted and come to dominate in its particular realm of physical, biological and chemical conditions. Our ability to predict initiation, maintenance, and dispersal of blooms on the Florida shelf has been severely limited by the lack of a quantitative description, or model, of their population dynamics and the physical, biological and chemical regime in which they are embedded. The modeling components of this project will incorporate the quantitative description of blooms and their surrounding environment provided by the field and laboratory portions of this project. The field component will employ a set of monthly hydrographic transects. Purpose: 1. Model the initiation, maintenance, and export of K. brevis red tides on the west Florida shelf at different time and space scales to predict landfall 2. Describe the physical habitat that effects transport and concentration of K. brevis 3. Determine the sources of inorganic and organic nutrients that allow growth and persistence of large K. brevis populations in coastal waters 4. Determine the production, occurrence, fate, and effects of brevetoxins in the environment during and after K. brevis blooms. ECOHAB (ECology and Oceanography of Harmful Algal Blooms) is a scientific program designed to increase understanding of the fundamental processes underlying the impacts and population dynamics of HABs. This program addresses the many factors at organismal, population, community, and ecosystem levels that determine how HAB species respond to, and potentially alter their environment, the manner in which HAB species affect food-web and community interactions. The program is based on the results of a workshop co-sponsored by National Oceanic and Atmospheric Administration (NOAA) and National Science Foundation (NSF) and currently receives funding from several federal partners including NOAA, NSF, the U.S. Navy and the U.S. Environmental Protection Agency (EPA). The entire sample is filtered through a C18 extraction disc to recover brevetoxins. The brevetoxins are eluted from the disc with 20 ml methanol under vacuum and reduced to 3 ml volume in preparation for analysis by high performance liquid chromatography (HPLC). Standard brevetoxins PbTx-2 and PbTx-3 are obtained from Chiral Corp. Miami, FL. Verification of PbTx-1 and PbTx-5 is accomplished by secondary standards recovered from K. brevis culture and identified by HPLC retention values. The amounts of intra- and extra-cellular toxins are determined in a 5-L sample processed through a stirred ultra-filtration cell concentrator (Amicon, Beverly MA). The sample was filtered through a 0.8 micron polycarbonate membrane (Osmonics, Westborough, MA) in the stirred cell at 5 psi to gently collect the viable K. brevis cells (intra-cell toxins) on the filter and to allow extra-cellular toxins to pass through with the filtrate. The Extra-Cellular toxins are recovered by passing the 4.8 L of filtrate through a C18 bonded-phase extraction disc as above. The Intra-cell toxins are extracted from approximately 200ml of solution remaining above the membrane (containing viable K. brevis cells) by rinsing the cell and membrane with distilled water and recovering toxins on a C18 extraction disc as above. Methanol extracts are reduced in volume to 3 ml in preparation for HPLC analysis as above. Methodology: Methodology Type: HPLC Pigments Methodology Description: Lab Analysis Methods Used: Photopigments: Photopigments are analyzed using high-performance liquid chromatography (HPLC). Aliquots of water were drawn under low vacuum onto GF/F glass-fiber filters that were immediately frozen and stored at -80 oC. At time of analysis, frozen filters were placed in 100% acetone, sonicated, and extracted in darkness at -20 oC for 12 to 16 hrs. Filtered extracts (up to 200 Error! Bookmark not defined.l) were injected directly into either 1) a Hewlett Packard (HP) model 1090 HPLC equipped with a single monomeric (ODS Hypersil; 100 x 4.6mm, 5 m) and two polymeric (250 x 4.6, 5 :m) reverse-phase C18 columns in series, a photodiode array detector, and a HP Model 1046A fluorometer placed in-line or 2) a Shimadzu model LC-XXX HPLC equipped with an ODS-Hypersil C18 column (200 x 4.6 mm, 4m particle size) and a diode array detector. The mobile phases and solvent flow rates followed that described by Pinckney et al. (1996) –for the HP – or Wright et al. (1991) – for the Shimadzu. Pigments were identified and quantified using authentic standards for chlorophylls and carotenoids. Absolute and relative phylogenetic group (and potentially species-specific) contributions to total Chl a biomass are derived from chemotaxonomic photopigments using ChemTax® software (Mackey et al 1996). Community Production: Bulk carbon fixation will be measured using 14C uptake experiments. Small volume samples (6-ml) inoculated with 14C are incubated in a temperature-controlled, variable-light photosynthetron (Prézelin et al. 1992) for 40 minutes. Samples are acidified in vials, transferred to the laboratory in darkness and and total activity later determined using a LKB model XXXX scintillation counter. From P-I measurements, non-linear fitting techniques will be used to determine the following P-I parameters; Pmax (maximum light saturated rate, a (light limited slope), Ik (minimum irradiance required to light-saturate photosynthesis) and b (slope at photoinhibiting irradiances) as described in Fahnenstiel et al. (1989).
